Biosecurity in Aquaculture

Numerous contaminations associated with the nodavirus are regularly reported worldwide in a great number of species of marine fish populating different ecosystems (platax and ombrine in Tahiti, grouper and bass of the Mediterranean Sea, sea bass of the Atlantic coasts), but also of other marine animals, such as the holothurian (sea cucumber) lagoon. These episodes of VNN (Viral Nervous Necrosis), also called Viral Encephalopathy and Retinopathy (VER), cause high mortalities and huge economic losses.

The emergence of these diseases and their expansion throughout the world is linked to the numerous exchanges of fishes and biological materials (broodstock, eggs and juveniles), necessary for the development of aquaculture.

Nodavirus infection causes nervous disorders in an increasing number of marine species : larvae and juveniles are the most susceptible to infection with a high mortality rate.

There are several methods of analysis of the virus, but the detection of nodavirus is now often carried out by the PCR technique, a technology that accelerated the process to detect the virus. However, the analysis methods require specialized laboratory equipment and qualified operators.

The Nodavirus kit: a rapid detection test

The nodavirus detection kit is based on the enzymatic amplification of a specific nodavirus sequence by PCR.

The PCR calls upon a set of primers which amplify a specific DNA fragment of different strains of Nodavirus. The visualization of the amplified products is achieved by a specific hybridization with homologous probes deposited at predefined positions on ready-to-use membranes (mini-array).

A colorimetric revelation step via a blue spot on the nylon membrane is then performed. The interpretation of the results visualized on the membrane (mini-array) can be done with the naked eye.
